TES, chemically known as N-tris(hydroxymethyl)methyl-2-aminoethanesulfonic acid, is one of the Good buffers developed by Norman Good and his colleagues in the 1960s. The goal of Good's research was to create a series of buffers that were biologically inert, highly soluble in water, and stable at a wide range of temperatures and pH levels, making them ideal for biological and biochemical research.

TES is a zwitterionic buffer with the molecular formula C6H15NO6S. It is particularly useful for maintaining pH values ​​in the range of 6.8 to 8.2, with a pKa of 7.4 at 25°C. This makes it suitable for experiments that require neutral to slightly alkaline pH values. TES is highly soluble in water and has excellent buffering capacity, which is essential for a variety of biological and biochemical applications.

TES is widely used in cell culture media to maintain a stable pH environment, which is essential for the growth and maintenance of cells. Its buffering capacity helps prevent pH fluctuations that can affect cell function and viability.

TES is used in a variety of biochemical assays, especially those involving enzymatic reactions, where a stable pH is essential. It ensures that the pH remains constant, resulting in more accurate and reproducible results.

In protein purification protocols such as ion exchange chromatography, TES provides the stable pH environment required for protein binding and elution. This stability helps maintain protein integrity and functionality throughout the purification process.

TES is used as a buffer in electrophoresis to separate proteins and nucleic acids. Its ability to maintain a consistent pH ensures proper migration of biomolecules, resulting in clear and precise separations.

TES offers several advantages, including minimal absorption in the UV and visible spectra, which reduces interference in optical measurements. Its chemical stability and resistance to enzymatic degradation ensure its effectiveness over extended periods of time and under a variety of experimental conditions. Additionally, TES does not form complexes with metal ions, making it ideal for studies involving metal-dependent enzymes.
