PyBOP was introduced in 1990 to retain BOP-like peptide-coupling performance while avoiding BOP's HMPA byproduct. Its phosphonium chemistry activates carboxylic acids for amide formation, while pyrrolidino substitution changes the waste chemistry. PyBOP marks an important shift in reagent design: success means considering not only the desired bond, but also what byproducts the reaction creates.
